Associate Scientist, Molecular Biology at CSL Seqirus

We have an exciting opportunity to join our Biopharmaceutical Product Development (BPD) department to assist sa-mRNA-based therapeutics development. You will play an important role in high throughput sa-mRNA production and testing, with a focus on molecular cloning to aid in construct generation. You will be an important contributor to a cross functional team and is expected collaborate with analytical and LNP process development teams.

You will be reporting to the Scientist II. You will work onsite at our CSL Seqirus site in Holly Springs, NC location. Responsibilities:

• Design and execute molecular cloning strategies to produce new plasmid sequences for sa-mRNA production, troubleshoot the cloning process as needed.
• Amplify plasmid in E. Coli cells and perform purification using state-of-the-art automation system.
• Conduct plate-based enzymatic digestion and in-vitro transcription reactions.
• High throughput purification of linear DNA and sa-mRNA.
• Run capillary electrophoresis to analyze the quality of RNA/DNA samples.
• Participate in the development, validation and scaling up of novel assays.
• Work with automated liquid handlers to enable high throughput sa-mRNA production.
• Maintain a comprehensive Electronic Lab Notebook (ELN) and ensure all experimental procedures and documentation are compliant with highest quality and regulatory standards.
• Use GraphPad Prism and other software for data analysis and figure generation.

Qualifications:

• BS or MS in Biological Sciences or a related field
• 4+ years of experience in pharmaceutical/biotechnology labs.
• Demonstrated proficiency in molecular biology techniques including but not limited to: RNA/DNA isolations such as miniprep/maxiprep, molecular cloning (Gibson assembly/Golden Gate), agarose gel electrophoresis, PCR, etc. is required.
• Experience with Geneious, Benchling, or SnapGene.
• Bacterial transformation, plating, culturing, DNA prep, and colony screening.
• Experience in nucleic acids handling, quantitation, and analysis (e.g. Nanodrop, Qubit, Fragment analyzer, RT-qPCR, ddPCR, etc.).
• Familiarity with Hamilton or Tecan.
